The SpotOn MinION Flow Cell (R10.4.1) is a consumable designed for use with MinION and GridION devices. It is optimized for high consensus accuracy, particularly when paired with V14 sequencing kits. The R10.4.1 nanopores feature a double reader-head, enabling accurate base calling with a modal raw read accuracy of over 99%.
Key Features
- High Accuracy with Double Reader-Head:
- R10.4.1 nanopores enhance sequencing accuracy, ideal for high-consensus applications.
- Paired with Kit 14 chemistry, it provides modal raw read accuracies above 99%.
- Extended Usage:
- Can run continuously for ~72 hours under optimal conditions.
- Allows for run-stop-wash workflows to reuse the flow cell for multiple libraries.
- Real-Time Data Streaming:
- Sequencing data is streamed in real time, enabling users to monitor progress and terminate runs as soon as sufficient data is collected.
- Multiplexing Capability:
- Compatible with ONT's barcoding kits, allowing up to 96 samples per run for cost-efficient sequencing.
- Environmentally Conscious Design:
- Includes a light shield for optimal performance.
- Packaging doubles as a returns kit for responsible recycling.
Workflow
- Preparation:
- Load the prepared DNA/RNA library onto the flow cell.
- Install the light shield and start the sequencing experiment.
- Sequencing:
- DNA is sequenced by the nanopores, and data is streamed in real time.
- Use MinKNOW software to monitor performance and analyze sequencing data.
- Flow Cell Checks:
- Perform a flow cell check before loading samples to verify the number of active nanopores.
- Flow cells with fewer than 800 nanopores (reported within 2 days of check) are eligible for replacement.
- Reuse:
- Use the Flow Cell Wash Kit to wash out libraries and prepare the flow cell for subsequent runs.
Storage and Stability
- Room Temperature: Store unopened flow cells at room temperature for up to 1 month.
- Refrigerated (2–8°C): Store unopened flow cells for up to 12 weeks.
- Warranty: 12 weeks from receipt; a flow cell check is required within this period.
