Scientific Background
The E7 proteins of HPV types 16 and 18 are critical for viral oncogenesis. These small (~15 kDa) proteins inactivate pRB, disrupting cell cycle regulation and contributing to uncontrolled cell proliferation. Structurally, E7 contains a zinc finger motif and an LXCXE sequence essential for pRB binding. The disordered N-terminal and structured C-terminal regions of the E7 protein are evolutionarily conserved and key to its transforming function. Persistent infection with HPV16 or HPV18 is associated with ~70% of cervical and anogenital cancers, as well as a growing number of HPV-related head and neck cancers.
