Basal Medium for the Cultivation of Aerobic and Anaerobic Bacteria
Liofilchem® Schaedler Broth is a highly enriched, non-selective liquid medium specifically designed to support the growth of both aerobic and anaerobic bacteria, including fastidious species. Its carefully balanced formulation provides essential nutrients such as peptones, glucose, yeast extract, hemin, and L-cystine to support optimal bacterial recovery. This broth is particularly valuable in clinical diagnostics, pharmaceutical microbiology, food safety testing, and environmental monitoring where broad microbial recovery under variable oxygen conditions is required.
Key Features
- Non-selective broth suitable for a wide range of bacterial isolates
- Optimized for both aerobic and anaerobic organisms, including fastidious anaerobes
- Hemin and L-cystine supplementation promotes growth of hemin-requiring organisms
- Free from thioglycollate to reduce inhibitory effects on sensitive anaerobes
- Suitable for use in anaerobic jars or chambers and various enrichment protocols
- Ideal for fecal, environmental, and pharmaceutical applications
Typical Formula (per liter)
- Tryptic Soy Broth – 10.0 g
- Tryptone – 5.0 g
- Yeast Extract – 5.0 g
- Glucose – 5.0 g
- Hemin – 0.01 g
- L-Cystine – 0.4 g
- Final pH: 7.6 ± 0.2 at 25 °C
Preparation Instructions
- Suspend 25.4 g of the dehydrated powder in 1 liter of distilled or deionized water.
- Heat to boiling until fully dissolved.
- Dispense into final containers.
- Sterilize by autoclaving at 121 °C for 15 minutes.
- If not used immediately, regenerate by heating to 100 °C for 20 minutes to re-establish anaerobic conditions (only once).
- Inoculate and incubate at 36 ± 1 °C for 18–48 hours under suitable atmospheric conditions.
Applications
- Cultivation of Clostridium spp., Streptococcus spp., Staphylococcus spp., Bacteroides spp., and Escherichia coli
- Studies on gastrointestinal or environmental microbiota
- Routine culture in pharmaceutical and clinical laboratories
- Enrichment prior to anaerobic plating or susceptibility testing
- Validation of microbial limits in raw materials and finished products
