The HEMO-ANAEROBIC CULTURING system is a liquid culturing medium based on Schaedler + Vitamin K1 Broth, specifically designed for isolating fastidious anaerobic and facultative anaerobic microorganisms from venous blood. It is available in three presentations to cater to different patient demographics:

  1. HEMO-ANAEROBIC CULTURING – For adults.
  2. HEMO-ANAEROBIC CULTURING Pediatric – For children.
  3. HEMO-ANAEROBIC CULTURING Neonatal – For newborns and children under 2 years.

Product Features

  • Nutritive Formula: Contains essential nutrients like tryptone, peptone, yeast extract, L-cystine, hemin, and Vitamin K1 to promote the growth of fastidious microorganisms.
  • Anticoagulant and Growth Enhancers: Sodium-polyanethol-sulphonate (SPS) neutralizes human serum’s bactericidal effects, inhibits phagocytosis, and partially inactivates specific antibiotics.
  • Sterile Packaging:
    • Adult and Pediatric Bottles: Red plastic caps with perforable gray rubber subcaps and transparent protective covers.
    • Neonatal Bottles: Perforable gray rubber stoppers with metallic screw caps and red liftable plastic overcaps.

Test Procedure

a) Inoculum Technique

  1. Inspect the bottle for contamination.
  2. Disinfect the external side of the gray rubber stopper.
  3. Inject the venous blood aseptically into the bottle.
    • Volumes:
      • Adults: 8–10 mL
      • Pediatrics: 4–5 mL
      • Neonates: 0.5–1 mL
  4. Close the protective cap and mix blood with the medium.
  5. Label the bottle and incubate at 36 ± 1°C.

b) Subculturing Technique

  1. Mix the bottle gently to suspend the culture medium.
  2. Inoculate selective plates with 1–2 drops of the medium for isolating Gram-positive and Gram-negative anaerobic microorganisms.
  3. Perform Gram staining and microscopic examination at 100X magnification.
  4. Repeat subculturing at 24, 48, and 72 hours. Extend incubation to 7 days if no microbial growth is observed before concluding results.

Limitations

  • Microorganisms might fail to grow due to:
    • Absence in the sample.
    • Inhibitory antibiotic concentrations.
    • Strain fastidiousness or insufficient incubation time.

Storage and Shelf Life

  • Storage: Store at 10–25°C in original packaging. Avoid heat and temperature fluctuations.
  • Shelf Life: Valid until the expiry date on the label. Discard if deterioration is evident.
