Usage Recommendations:
- Sample Preparation:
- Polarize human peripheral blood CD4+ T cells under Th17 conditions.
- Stimulate cells using appropriate reagents (e.g., Cell Stimulation Cocktail).
- Staining Protocol:
- Use the Intracellular Fixation & Permeabilization Buffer Set for effective staining.
- Add 5 µL (0.06 µg) of antibody per 100 µL of cell suspension.
- Flow Cytometry:
- Analyze samples using a flow cytometer equipped with a red laser (633–647 nm).
- Fixation:
- Fix samples in IC Fixation Buffer to preserve fluorescence for up to 3 days.
Storage and Handling:
- Store at 4°C in the dark to protect the APC fluorochrome.
