    The Invitrogen™ ImaGene Green™ C12FDG lacZ Gene Expression Kit provides a high-sensitivity, fluorescence-based system for detecting β-galactosidase (lacZ) reporter gene expression in live cells. The kit’s core substrate, C12FDG (5-Dodecanoylaminofluorescein Di-β-D-Galactopyranoside), is a fluorescein-based β-galactosidase substrate covalently modified with a 12-carbon lipophilic tail to enhance cellular retention of the fluorescent product.

    Once inside the cell, C12FDG is cleaved by β-galactosidase to produce a bright green fluorescent product (excitation/emission ~490/514 nm) that is stably retained, likely via incorporation of its lipophilic moiety into cellular membranes. The kit also includes PETG, a broad-spectrum β-galactosidase inhibitor, and chloroquine diphosphate to prevent acidic hydrolysis, ensuring specific and accurate reporter detection.
