Assay Principle
The ELISA plate is pre-coated with a monoclonal antibody specific to MDA. Standards or test samples are added and bind to the immobilized antibody. An HRP-conjugated detection antibody is then added, forming a sandwich complex. TMB substrate is introduced and converted to a blue product, which turns yellow upon the addition of stop solution. Absorbance is read at 450 nm, and MDA concentration is calculated using a standard curve.

Assay Procedure Summary
- Standard Dilution: Prepare six standards (540 → 30 ng/mL).
- Add 40 μL sample diluent + 10 μL sample to wells (except blank).
- Incubate 30 minutes at 37°C, sealed.
- Wash plate 5× using diluted wash buffer.
- Add 50 μL HRP-conjugate, incubate again.
- Wash and add 50 μL each of Chromogen A & B.
- Incubate 15 minutes in the dark at 37°C.
- Add 50 μL stop solution. Measure OD at 450 nm within 15 minutes.
