Intended Use

Assay Principle
This kit employs a Sandwich ELISA method:
- The pre-coated microplate wells contain immobilized Hepatitis B surface antigen (HBsAg).
- Samples and standards containing HBsAb are added and bind to the coated antigen.
- A horseradish peroxidase (HRP)-conjugated HBsAg is added to form a sandwich complex with bound HBsAb.
- After washing to remove unbound components, TMB substrate is added and color develops.
- The reaction is stopped and optical density is measured at 450 nm.
- OD values are proportional to the concentration of HBs-Ab in the sample.

Sample Preparation Guidelines
- Serum/Plasma: Centrifuge at 2,000–3,000 rpm for 20 minutes. Avoid hemolysis and precipitates.
- Cell culture supernatants: Centrifuge at 2,000–3,000 rpm for 20 minutes.
- Cell lysates: Use PBS (pH 7.2–7.4), subject to repeated freeze-thaw cycles, and centrifuge.
- Tissue homogenates: Homogenize tissue in PBS, centrifuge, and collect supernatant.
- Storage: Store samples at –20°C if not assayed immediately. Avoid repeated freeze-thaw cycles.
Sodium azide (NaN₃) should not be present in samples as it inhibits HRP enzyme activity.
Assay Procedure (Summary)
- Prepare a standard curve using serial dilutions: 135, 90, 60, 30, 15, and 7.5 IU/L.
- Add 50 µL of standard/sample + 40 µL sample dilution buffer into each well.
- Incubate at 37°C for 30 min, then wash 5 times with diluted wash buffer.
- Add 50 µL of HRP-conjugate reagent, incubate again for 30 min at 37°C.
- Wash as before and add 50 µL of Chromogen A and B to each well.
- Incubate in the dark at 37°C for 15 min.
- Add 50 µL of Stop Solution to terminate the reaction.
- Read absorbance at 450 nm within 15 minutes.

Important Notes
- All reagents should be brought to room temperature before use.
- Avoid light exposure during color development (TMB reaction).
