The Gibco™ TrypLE™ Express Enzyme (1X), Phenol Red, is a recombinant, animal origin-free (AOF) enzyme formulated for gentle and efficient dissociation of adherent mammalian cells. This enzyme is a direct replacement for trypsin and offers superior performance, reduced cell damage, and enhanced convenience for a wide range of cell culture applications.
Key Features:
- Gentle on Cells:
- Cleaves peptide bonds at the C-terminal sides of lysine and arginine with minimal damage, ensuring higher cell viability.
- Room-Temperature Stable:
- Stable for up to 24 months at room temperature (15–30°C), eliminating cold storage challenges.
- Phenol Red Indicator:
- Contains phenol red for pH monitoring during cell dissociation procedures.
- Animal Origin-Free (AOF):
- Free from animal-derived components, making it suitable for regulatory-compliant workflows and sensitive applications.
- Ready-to-Use:
- 1X liquid formulation allows direct use in standard protocols without dilution or additional preparation.
- Self-Inactivating:
- Dilution inactivates TrypLE Express, eliminating the need for trypsin inhibitors like FBS.

- Primary Tissue Dissociation:
- Ideal for breaking down tissues into single-cell suspensions for downstream applications.
- Cell Passaging:
- Provides a gentle solution for routine subculturing of sensitive adherent cells.
- Phenol Red-Dependent Assays:
- Suitable for workflows requiring a pH indicator for monitoring during dissociation.
Advantages:
- High Purity and Specificity:
- Reduces non-specific cleavage and potential damage caused by multiple enzymes in traditional trypsin.
- Regulatory Compliance:
- Manufactured under cGMP conditions at ISO 13485-certified facilities.
- Easy Substitution:
- Seamlessly replaces trypsin in existing protocols with no changes required.
- Enhanced Workflow Efficiency:
- Ready-to-use formulation minimizes preparation time and simplifies handling.
- Eco-Friendly Manufacturing:
- Produced using energy-efficient methods and sustainable packaging.
Protocol Overview:
- Pre-Warm TrypLE:
- Warm TrypLE Express to room temperature before use.
- Prepare Cells:
- Wash adherent cells with PBS to remove residual serum, which can inhibit the enzyme.
- Apply TrypLE:
- Add enough TrypLE Express to cover the cell monolayer.
- Incubate:
- Incubate at 37°C for 2–5 minutes, monitoring for cell detachment.
- Neutralize:
- Dilute with complete growth medium to inactivate TrypLE Express.
- Centrifuge and Resuspend:
- Centrifuge the cells and resuspend in fresh medium for further processing.
