The Gibco™ TrypLE™ Express Enzyme (1X), Phenol Red, is a recombinant, animal origin-free (AOF) enzyme formulated for gentle and efficient dissociation of adherent mammalian cells. This enzyme is a direct replacement for trypsin and offers superior performance, reduced cell damage, and enhanced convenience for a wide range of cell culture applications.

Key Features:

  1. Gentle on Cells:
    • Cleaves peptide bonds at the C-terminal sides of lysine and arginine with minimal damage, ensuring higher cell viability.
  2. Room-Temperature Stable:
    • Stable for up to 24 months at room temperature (15–30°C), eliminating cold storage challenges.
  3. Phenol Red Indicator:
    • Contains phenol red for pH monitoring during cell dissociation procedures.
  4. Animal Origin-Free (AOF):
    • Free from animal-derived components, making it suitable for regulatory-compliant workflows and sensitive applications.
  5. Ready-to-Use:
    • 1X liquid formulation allows direct use in standard protocols without dilution or additional preparation.
  6. Self-Inactivating:
    • Dilution inactivates TrypLE Express, eliminating the need for trypsin inhibitors like FBS.

Applications:

  1. Adherent Mammalian Cell Culture:
    • Efficiently dissociates various cell types, including CHO, HEK 293, A549, keratinocytes, and embryonic stem cells.
  2. Primary Tissue Dissociation:
    • Ideal for breaking down tissues into single-cell suspensions for downstream applications.
  3. Cell Passaging:
    • Provides a gentle solution for routine subculturing of sensitive adherent cells.
  4. Phenol Red-Dependent Assays:
    • Suitable for workflows requiring a pH indicator for monitoring during dissociation.

Advantages:

  1. High Purity and Specificity:
    • Reduces non-specific cleavage and potential damage caused by multiple enzymes in traditional trypsin.
  2. Regulatory Compliance:
    • Manufactured under cGMP conditions at ISO 13485-certified facilities.
  3. Easy Substitution:
    • Seamlessly replaces trypsin in existing protocols with no changes required.
  4. Enhanced Workflow Efficiency:
    • Ready-to-use formulation minimizes preparation time and simplifies handling.
  5. Eco-Friendly Manufacturing:
    • Produced using energy-efficient methods and sustainable packaging.

Protocol Overview:

  1. Pre-Warm TrypLE:
    • Warm TrypLE Express to room temperature before use.
  2. Prepare Cells:
    • Wash adherent cells with PBS to remove residual serum, which can inhibit the enzyme.
  3. Apply TrypLE:
    • Add enough TrypLE Express to cover the cell monolayer.
  4. Incubate:
    • Incubate at 37°C for 2–5 minutes, monitoring for cell detachment.
  5. Neutralize:
    • Dilute with complete growth medium to inactivate TrypLE Express.
  6. Centrifuge and Resuspend:
    • Centrifuge the cells and resuspend in fresh medium for further processing.
