Principle of the Assay
The Human Hepatitis B Virus Surface Antigen (HBsAg) ELISA Kit is based on a solid-phase sandwich enzyme-linked immunosorbent assay (ELISA).
1️⃣ Capture Antibody Coating – The microplate wells are pre-coated with a target-specific antibody that binds to HBsAg in the sample.
2️⃣ Sample Addition & Incubation – The test sample is added to the wells, and HBsAg binds to the immobilized antibody.
3️⃣ Detection Antibody Binding – A biotinylated secondary antibody binds to a different epitope of HBsAg, forming a sandwich complex.
4️⃣ HRP Conjugate & Signal Generation – HRP-conjugated streptavidin is added, followed by TMB substrate, producing a colorimetric signal.
5️⃣ Stop Reaction & Read Absorbance – The reaction is stopped using Stop Solution, and absorbance is measured at 450 nm using a microplate reader.
The intensity of the color is proportional to the concentration of HBsAg in the sample.
