Liofilchem® Voriconazole (VO) Antimicrobial Susceptibility Disc (1 µg) is a high-quality, ready-to-use filter paper disc impregnated with 1 microgram of voriconazole, a second-generation triazole antifungal agent used in the treatment of serious systemic fungal infections. Designed for in vitro antifungal susceptibility testing (AFST) using the Kirby-Bauer disk diffusion method, this disc is optimized for evaluating the activity of voriconazole against yeasts and filamentous fungi, including Candida and Aspergillus species.

Manufactured in accordance with ISO 9001 and ISO 13485 quality management standards, the Liofilchem® Voriconazole disc is suitable for use in clinical diagnostic laboratories, resistance surveillance programs, and pharmaceutical R&D.

    Key Features

    • Precise potency: Each disc delivers 1 µg of voriconazole, ensuring standardized and reproducible inhibition zones.
    • Broad-spectrum antifungal activity: Highly effective against Candida albicans, Candida glabrata, Aspergillus fumigatus, and other medically important fungi.
    • Critical for azole resistance detection: Particularly relevant for detecting reduced susceptibility or resistance in Candida auris, C. krusei, and azole-resistant Aspergillus isolates.
    • CLSI and EUCAST compatible: Validated for use with standardized zone diameter breakpoints for Candida spp. and filamentous fungi.
    • Sterile and stable: Supplied in sealed cartridges (50 discs) or canisters (250 discs) with desiccant to preserve antifungal potency.
    • Color-coded labeling: Facilitates quick identification in multi-disc workflows.

    Applications and Use Cases

    1. Clinical Mycology

    • AFST of Candida spp., especially in bloodstream infections, mucosal candidiasis, and deep-seated infections.
    • Used to test Aspergillus spp. in cases of invasive pulmonary aspergillosis and chronic pulmonary aspergillosis.
    • Monitor susceptibility in Candida auris and Aspergillus fumigatus to detect emerging resistance.

    2. Antifungal Resistance Surveillance

    • Track azole resistance patterns in healthcare and community-acquired fungal infections.
    • Support global antifungal stewardship initiatives.

    3. Pharmaceutical and Research Use

    • Evaluate voriconazole activity in comparison with other azoles (e.g., fluconazole, itraconazole, posaconazole).
    • Study mutations in ERG11, FKS, and other genes conferring azole resistance.

    4. Veterinary and Environmental Mycology

    • Assess fungal susceptibility in veterinary infections and environmental fungal isolates relevant to public health.

    Use Instructions

    1. Medium: Use RPMI 1640 agar buffered with MOPS (recommended for fungi), or other validated fungal susceptibility testing media.
    2. Inoculum Preparation:
      • Yeasts: Adjust to 0.5 McFarland standard.
      • Moulds: Prepare standardized conidial suspensions (per CLSI M38 or EUCAST protocols).
    3. Inoculation: Evenly swab inoculum over the entire agar surface using a sterile swab.
    4. Disc Placement: Place one 1 µg Voriconazole disc on the agar using sterile forceps or a disc dispenser.
    5. Incubation:
      • Yeasts: 35 ± 2 °C for 24–48 hours
      • Moulds: 35 °C for 48–72 hours
    6. Interpretation:
      • Measure the zone of inhibition (in mm) and interpret according to CLSI M44 / M61 or EUCAST AFST guidelines.
      • Categorize isolates as Susceptible (S), Intermediate (I), or Resistant (R).
