The Liofilchem TERGITOL 1.5% Supplement is a sterile liquid additive used in conjunction with Tergitol Agar Base (ref. 610184 / 620184). When incorporated into the agar base, this supplement creates a highly selective medium for the detection, enumeration, and identification of Escherichia coli and coliform bacteria in water samples. Its inhibitory properties, combined with Tergitol NP-7, promote the recovery of coliforms by suppressing Gram-positive bacteria, spore-forming Gram-negative bacteria, and Proteus spp. swarming.

Preparation & Usage Instructions
- Preparation:
- Aseptically add 7 mL of TERGITOL 1.5% Supplement to 1 litre of autoclaved and cooled (45-50°C) Tergitol Agar Base.
- Mix thoroughly to ensure even distribution of the supplement.
- Additional Supplementation:
- Add 2.5 mL of TTC 1% Supplement (ref. 80300 / 80430) to 1 litre of base medium after sterilization.
- Dispensing:
- Pour the prepared medium into sterile Petri dishes or final containers.
- Inoculation:
- Use membrane filtration or direct plating techniques to inoculate the medium.
- Incubation:
- For total coliforms: Incubate at 37°C for 24 hours.
- For E. coli: Incubate at 44°C for 16-24 hours.
Interpreting Results
- Typical Colony Morphology:
- Escherichia coli: Yellow-orange colonies on a yellow medium.
- Enterobacter/Klebsiella: Yellow colonies on a yellow medium.
- Proteus/Salmonella/Shigella: Dark red colonies on a green medium.
- Use further confirmatory tests, if necessary, for precise identification.
