The INTEGRAL SYSTEM GARDNERELLA is an 18-well system designed for the biochemical identification and susceptibility testing of Gardnerella vaginalis. It uses colorimetric changes in culture media containing desiccated biochemical substrates to identify the bacterium and assess its susceptibility to antibiotics. Clinical samples such as vaginal swabs, urethral secretions, or seminal fluids can be analyzed with this system. Incubation is carried out at 36 °C ± 1 °C for 18-24 hours.

Package Contents

  • 20 INTEGRAL SYSTEM GARDNERELLA systems
  • 20 vials of Physiological Solution (7.0 ml/vial)
  • 1 bottle of Ninhydrin Reagent (5.0 ml)
  • 1 bottle of H2O2 Reagent (5.0 ml)
  • 1 amber dropper bottle for Ninhydrin Reagent
  • Instruction sheet

Additional Items Required (Not Included)

  • Vaseline oil for microbiological use (2 bottles of 50 ml, cod. 80278)
  • Standard microbiology laboratory materials

System Configuration

The system comprises two groups of tests:

  1. Biochemical Identification (Wells 1-12)
Includes biochemical substrates for hydrolysis, fermentation, and enzymatic reactions.
  2. Susceptibility Testing (Wells 13-18)
Tests for antibiotic sensitivity against G. vaginalis.

17-SXTMedium with Co-Trimoxazole (30 μg/ml)Susceptibility Testing
18-CControl well (medium without antibiotics)Growth Control

Test Procedure

  1. Preparation:
    • Take a system from the kit.
    • Prepare a bacterial suspension in the provided Physiological Solution (0.5-1.0 McFarland turbidity).
  2. Inoculation:
    • Dispense 0.2 ml of the bacterial suspension into each well.
    • Add 0.1 ml of the suspension to well 1-IPP.
    • Cover well 6-UR with 2 drops of sterile vaseline oil.
    • Cover the system and incubate at 36 °C ± 1 °C for 18-24 hours.
  3. Reagent Addition (Post-Incubation):
    • Add 2 drops of Ninhydrin Reagent to well 1-IPP.
    • Add 2 drops of H2O2 Reagent to well 5-CAT.
  4. Result Interpretation:
    • Observe color changes in wells 1-12 (biochemical identification) and 13-17 (susceptibility testing).
    • Ensure the control well (18-C) turns yellow, indicating successful growth.

Quality Control

Each batch is tested using Gardnerella vaginalis ATCC 14018. Negative control involves using Physiological Solution without bacteria.

Storage

  • Store at 2-8 °C in the original packaging.
  • Avoid exposure to heat and temperature fluctuations.
  • Discard if signs of deterioration are evident.

Precautions

  • INTEGRAL SYSTEM GARDNERELLA is intended for in vitro diagnostic use only.
  • Use in a laboratory setting by trained personnel under aseptic conditions.
