Liofilchem® Motility Test Agar is a dehydrated culture medium formulated for the detection of motility in gram-negative enteric bacilli. It complies with the UNI EN ISO 11290-1:2005 standard and is widely used in clinical and food microbiology laboratories. This low-concentration agar formulation enables visible detection of motile bacterial species via the diffusion of growth from the stab line in tube inoculations.
Key Features
- Standardized Composition: Contains casein peptone and meat extract to provide essential nutrients, with a reduced agar content (3.5 g/L) to support motility detection.
- Validated Method: Developed according to ISO 11290-1:2005 guidelines for routine testing of gram-negative enteric bacteria.
- Clear Visualization: Low agar concentration creates a semi-solid matrix allowing motile bacteria to spread while non-motile bacteria remain along the stab line.
- Versatile Packaging: Available in 100 g and 500 g plastic bottles for flexible lab-scale preparation.
- Extended Shelf Life: Up to 4 years when stored under recommended conditions.
Typical Composition (per liter)
Component | Concentration |
---|---|
Casein Peptone | 20.0 g/L |
Meat Extract | 6.1 g/L |
Agar | 3.5 g/L |
Final pH | 7.2 ± 0.2 |
Applications and Use Cases
1. Clinical Microbiology
- Differentiation of motile and non-motile Enterobacteriaceae, e.g., Escherichia coli (motile) vs Klebsiella pneumoniae (non-motile).
2. Food and Water Testing
- Detection of motility as part of Listeria spp. differentiation workflow, especially in food safety labs.
3. Educational Use
- Demonstrating microbial motility in academic settings and microbiology training programs.
Use Instructions
Preparation:
- Suspend 29.6 g of powder in 1 L of distilled or deionized water.
- Heat to boiling with gentle shaking until completely dissolved.
- Dispense into final containers.
- Sterilize by autoclaving at 121°C for 15 minutes.
Inoculation:
- Inoculate by vertically stabbing a pure culture into the medium to more than half the depth of the tube.
Incubation:
- Primary: 36 ± 1°C for 24–48 hours in aerobic conditions.
- Optional: Reincubate negative tubes at 25 ± 2°C for up to 5 days.
Interpretation:
- Motile organisms: Show diffuse or hazy growth radiating from the stab line.
- Non-motile organisms: Growth confined to the stab line only.
