Usage Recommendations:

1. Sample Preparation:
   • Stimulate human peripheral blood cells under Th17-polarizing conditions using IL-23 or similar reagents.
2. Staining Protocol:
   • Use the Intracellular Fixation & Permeabilization Buffer Set for intracellular staining.
   • Add 5 µL (0.5 µg) of antibody per 100 µL of cell suspension.
3. Flow Cytometry:
   • Analyze samples using a flow cytometer equipped with a violet laser (405 nm).
4. Fixation:
   • Fix samples in appropriate buffers to preserve fluorescence for up to 3 days.

Storage and Handling:

• Store at 4°C in the dark to protect the eFluor™ 450 fluorochrome.
