Liofilchem® Brain Heart Infusion (BHI) Broth is a highly nutritious, general-purpose liquid medium designed for the cultivation of a wide range of aerobic and anaerobic bacteria, including fastidious microorganisms from clinical, food, water, and environmental samples. It is particularly suitable for the plasma coagulase test used to identify coagulase-positive staphylococci, as prescribed by ISO 6888 standards.

The formulation also complies with recommendations from the Clinical and Laboratory Standards Institute (CLSI) for the preparation of inocula in antimicrobial susceptibility testing and from the American Public Health Association (APHA) for microbiological analysis of water and wastewater.

Preparation Instructions (Dehydrated Medium)

  1. Suspend 37 g of BHI powder in 1 L of distilled or deionized water.
  2. Mix thoroughly and heat to boiling, shaking frequently until completely dissolved.
  3. Dispense into final containers.
  4. Autoclave at 121 °C for 15 minutes.
  5. (Optional) Reduce media for anaerobic use by:
    • Incubating under anaerobic conditions for 18–24 h, or
    • Heating to 100 °C in a boiling water bath, with caps loosened during heating and tightened during cooling.

Test Procedures

1. General Cultivation

  • Inoculate sample into BHI Broth directly or after plate inoculation.
  • Incubate aerobically at 37 ± 1 °C for 24–72 hours.
  • Observe for turbidity, which indicates microbial growth.

2. Coagulase Test (ISO 6888)

  • Inoculate BHI Broth with a colony from Baird-Parker Agar.
  • Incubate at 37 ± 1 °C for 24 ± 2 hours.
  • Mix 0.1 mL of culture with 0.3 mL rabbit plasma.
  • Incubate at 37 °C for 4–6 hours and assess plasma clotting.
  • A clot larger than half the total volume indicates a positive result.
