    Liofilchem® Campylobacter Blood Free Medium Base is a selective, blood-free, charcoal-based medium used with Campylobacter CCDA Supplement (ref. 81037) for the isolation and enumeration of thermotolerant Campylobacter species, including C. jejuni, C. coli, and C. lari, from food, environmental, and clinical samples.

    This medium, also known as modified Charcoal Cefoperazone Deoxycholate Agar (mCCDA), conforms to ISO 10272 and EN ISO 11133 standards, and is designed to replace blood-containing formulations with charcoal, ferrous sulfate, and sodium pyruvate. It provides improved selectivity and reliability in detecting Campylobacter spp. in compliance with international food safety and public health testing protocols.

    Principle of the Method

    • Nutrient-rich components (meat extract, tissue digest, and casein hydrolysate) supply essential growth factors for Campylobacter.
    • Charcoal acts as a detoxifying agent by absorbing toxic metabolites.
    • Sodium deoxycholate inhibits most Gram-positive bacteria.
    • Ferrous sulfate and sodium pyruvate serve as oxygen scavengers, maintaining microaerobic conditions.
    • Agar provides the solid medium for colony formation.
    • Supplementation with Campylobacter CCDA Supplement (Cefoperazone + Amphotericin B) increases selectivity by suppressing accompanying flora.

    Preparation

    1. Suspend 45.5 g of powder in 1 L of deionized or distilled water.
    2. Heat and stir until fully dissolved.
    3. Autoclave at 121°C for 15 minutes.
    4. Cool to 45–50°C.
    5. Aseptically add 2 vials (10 mL total) of rehydrated Campylobacter CCDA Supplement (ref. 81037).
    6. Mix thoroughly and pour into sterile Petri dishes.

    Technique

    1. Inoculate plates by directly spreading the sample on the agar surface or after enrichment in Bolton Broth (ref. 470340), as per ISO 10272.
    2. Incubate at 41.5°C for 40–48 hours under microaerophilic conditions (5–6% O₂, 3–10% CO₂, 84–85% N₂).
    3. Examine for typical Campylobacter colonies—greyish, flat, moist with metallic sheen and spreading edges.
    4. Confirm suspected colonies on Columbia Blood Agar (ref. 11025) by morphology, motility, oxidase, and temperature tolerance tests.
