Principle of the Assay
The assay is based on a two-step indirect ELISA method:
- Zika virus antigens coated on microplate wells bind specific antibodies present in the sample
- Unbound components are removed by washing
- Enzyme-labelled anti-human immunoglobulin binds to the antigen–antibody complex
- Addition of TMB substrate generates a blue color, which turns yellow after stop solution
- Color intensity correlates with the presence of Zika-specific antibodies
