Principle of the Assay
The assay is based on a two-step indirect ELISA method:
- Antigen-coated wells bind Chikungunya-specific antibodies present in the sample
- Unbound components are washed away
- Enzyme-labelled anti-human immunoglobulin binds to the antigen–antibody complex
- Addition of TMB substrate produces a blue color, which turns yellow upon stopping
- Color intensity correlates with the presence of antibodies in the sample
