Liofilchem™ G.N. Hajna Broth is a selective enrichment medium formulated for the cultivation and isolation of Gram-negative microorganisms, particularly Salmonella and Shigella, from clinical and non-clinical specimens. The medium contains a balanced mixture of nutrients, phosphate buffer, and selective agents that suppress Gram-positive flora and favor the growth of enteric pathogens.
The higher mannitol-to-glucose ratio and presence of sodium desoxycholate help maintain inhibitory conditions for normal fecal flora, while promoting early-phase growth of Shigella and Salmonella species.

Preparation
- Suspend 39.0 g of powder in 1 L of distilled or deionized water.
- Heat to boiling and shake until completely dissolved.
- Distribute into tubes.
- Sterilize by autoclaving at 121 °C for 15 minutes.
Technique
- Inoculate 1 g of feces or 1 mL of liquid stool into the broth, or insert swab specimens directly.
- Emulsify the specimen thoroughly.
- Incubate aerobically at 36 ± 1 °C for 6–8 hours.
- Transfer 1–2 drops of incubated broth onto selective plate media.
- Incubate plates aerobically at 36 ± 1 °C for 18–24 hours, then examine for pathogens.

Prepared medium: Light amber, slightly opalescent, without precipitate.
Incubation conditions: 36 ± 1 °C for 24 hours.
Storage
- Dehydrated medium: Store at 10–30 °C in a dry environment, tightly closed.
- Prepared tubes: Store at 2–8 °C.
- Use before expiry date or if no signs of deterioration or contamination are present.
