Preparation and Use
- Use at 1–2% concentration (10–20 g/L) for standard solid media preparation.
- For motility or semi-solid media, use 0.05–0.5% agar concentration.
- Dissolve in distilled or deionized water by heating, and sterilize by autoclaving (121°C, 15 min).
- Once solidified (~35°C), agar remains stable at typical incubation temperatures (37°C).
- Ensures uniform diffusion and proper oxygen gradients, facilitating growth of fastidious and anaerobic organisms.
Storage and Stability
- Store at 10–30°C, protected from moisture.
- Keep container tightly closed when not in use.
- Avoid extreme temperature fluctuations.
- Use before the expiry date indicated on the label or if deterioration is not evident (e.g., caking, discoloration).
