The Invitrogen™ eBioscience™ Protein Transport Inhibitor Cocktail (500X) is a ready-to-use, highly concentrated inhibitor formulation containing Brefeldin A and Monensin, two well-established agents that block intracellular protein transport. The cocktail is designed to prevent secretion of newly synthesized proteins, enabling their accumulation within the endoplasmic reticulum (ER) and Golgi apparatus.
This product is widely used in flow cytometry–based intracellular cytokine staining (ICS), functional immune assays, and protein expression studies, where detection of intracellularly retained proteins is required.
Mechanism of Action
- Brefeldin A disrupts protein transport from the ER to the Golgi by inhibiting COPI-mediated vesicle formation.
- Monensin, an ionophore, interferes with Golgi function by altering intracellular ion gradients.
Together, these inhibitors block the secretory pathway, leading to intracellular accumulation of cytokines, chemokines, and other secreted proteins. This accumulation allows sensitive downstream detection using intracellular staining, flow cytometry, or Western blotting.
