The MCE™ NAD⁺/NADH Assay Kit (WST-8) is a sensitive and convenient colorimetric assay designed to quantitatively detect both the oxidized (NAD⁺) and reduced (NADH) forms of coenzyme I in cells, tissues, and other biological samples. The assay is based on the WST-8 color reaction, enabling direct measurement without the need for tedious purification of NAD⁺/NADH from samples.
This kit is well suited for studies in cellular metabolism, mitochondrial function, oxidative stress, and redox balance, offering reliable and reproducible results with minimal sample handling.
Assay Principle
- NADH reduces the WST-8 reagent via an enzymatic reaction, generating a yellow-colored formazan product.
- The intensity of the color is directly proportional to the NADH concentration and is measured at 450 nm.
- NAD⁺ content is calculated indirectly by combining total NAD⁺/NADH measurement with selective NADH detection.
- The assay enables determination of total NAD⁺ + NADH, individual NADH levels, and the NAD⁺/NADH ratio.
