Liofilchem® Tryptose Phosphate Broth is a nutrient-rich liquid culture medium formulated for the growth and maintenance of fastidious microorganisms from clinical and environmental specimens.
It supports a wide range of bacteria, including Streptococcus spp., Neisseria spp., and other nutritionally demanding organisms.
This medium provides excellent recovery and proliferation of fastidious species and is compliant with international microbiological standards for clinical and sanitary testing.
Key Features
- Broad-spectrum growth support: Promotes the cultivation of Streptococcus pneumoniae, Neisseria meningitidis, Staphylococcus aureus, and Escherichia coli.
- High nutritional content: Enzymatic digest of casein provides amino acids, nitrogen, and carbon sources essential for microbial growth.
- Buffered system: Disodium phosphate maintains a stable pH (7.3 ± 0.2 at 25°C) throughout incubation.
- Energy source: Glucose supports rapid bacterial metabolism and growth.
- Osmotic balance: Sodium chloride maintains osmotic equilibrium for sensitive strains.
- Optional semi-solid modification: Addition of 1–2 g/L agar supports anaerobic growth in the lower zone of the medium.
- Excellent clarity: Ready-to-use medium appears clear to slightly opalescent, light amber in color.
Principle
The enzymatic digest of casein provides a complete range of nutrients—amino acids, vitamins, and minerals—while glucose serves as an energy source. Sodium chloride maintains osmotic stability, and disodium phosphate acts as a buffering agent to maintain a neutral environment conducive to microbial growth.
Typical Formula (g/L)
| Component | Concentration (g/L) |
|---|---|
| Enzymatic Digest of Casein | 20.0 |
| Glucose | 2.0 |
| Sodium Chloride | 5.0 |
| Disodium Phosphate | 2.5 |
| Final pH at 25°C: | 7.3 ± 0.2 |
Preparation
- Suspend 29.5 g of powder in 1 L of deionized or distilled water.
- Heat and stir until completely dissolved.
- Dispense into suitable tubes or flasks.
- Sterilize by autoclaving at 121°C for 15 minutes.
- For enhanced anaerobic conditions, add 1–2 g/L of agar.
Directions for Use
- Inoculate the broth with the specimen.
- Incubate at 35 ± 2°C for 18–48 hours.
- Observe for turbidity, indicating microbial growth.
