The Invitrogen™ eBioscience™ IHC Antigen Retrieval Solution (10X) is specially formulated for use in heat-induced epitope retrieval (HIER) during immunohistochemistry (IHC) of formalin-fixed, paraffin-embedded (FFPE) tissue sections. Formalin fixation can mask or alter protein epitopes, reducing antibody binding efficiency. This retrieval buffer, used in combination with controlled heat sources such as microwaves, water baths, or pressure cookers, helps restore antigenicity and improves staining sensitivity.
Supplied as a 10X concentrated stock, the solution must be diluted to 1X with reagent-grade water prior to use. It is available in two formulations to support different antibody requirements:
- High pH (pH 9.0): Enhances retrieval of certain nuclear and membrane proteins.
- Low pH (pH 6.0): Suitable for antigens more effectively unmasked under acidic conditions.
