Principle of Separation
Ficoll-Paque™ PLUS operates on the principle of density gradient centrifugation. When diluted whole blood or buffy coat is carefully layered onto the medium and centrifuged:
- Erythrocytes and granulocytes (higher density) migrate through the medium and pellet at the bottom
- Mononuclear cells (PBMCs) collect at the plasma–Ficoll interface
- Plasma and platelets remain above the gradient
