Liofilchem® Campylobacter CCDA Supplement is a selective antibiotic additive used to enrich Campylobacter Blood Free Medium Base (ref. 610130 / 620130) for the isolation and enumeration of thermotolerant Campylobacter species such as C. jejuni, C. coli, and C. lari from food, clinical, and environmental samples.

Based on the Bolton–Hutchinson formulation (1984), this supplement provides enhanced selectivity and recovery efficiency. It replaces cefazolin from the original medium with cefoperazone, improving isolation rates and specificity. The addition of amphotericin B ensures inhibition of fungal contaminants, allowing optimal Campylobacter growth under microaerophilic conditions.

Composition

Principle of the Method

Campylobacter spp. are fastidious, microaerophilic bacteria requiring specialized media for selective isolation.

• Cefoperazone replaces cefazolin in the traditional formulation, enhancing selectivity and inhibiting competing enteric flora.
• Amphotericin B suppresses fungal growth, improving isolation purity.
• The supplement enables selective recovery of Campylobacter species when combined with Campylobacter Blood Free Medium Base and incubation under microaerophilic conditions (5–10% O₂).
• Studies show higher isolation rates when incubating plates at 37 °C rather than 42 °C, providing flexibility in testing.

Preparation and Use

1. Aseptically reconstitute one vial of Campylobacter CCDA Supplement with 5 mL of sterile distilled water.
2. Mix gently until completely dissolved, avoiding foam formation.
3. Add the entire reconstituted solution to 500 mL of autoclaved Campylobacter Blood Free Medium Base cooled to 45–50 °C.
4. Mix thoroughly and pour into sterile Petri dishes.
5. Incubate inoculated plates at 36 ± 1 °C for 48 hours under microaerophilic conditions.

Storage and Stability

• Store at 2–8 °C in the original sealed container.
