Selective supplement for isolation of Legionella spp. (ISO 11731 compliant)

The Liofilchem® Legionella GVPC Supplement is a lyophilized selective additive formulated for use with Legionella Growth Supplement (Ref. 80056) and Legionella BCYE Agar Base (Refs. 610049, 620049). Together, these components create a selective Buffered Charcoal Yeast Extract (BCYE) medium that supports the growth of Legionella species while suppressing competing background flora.

This supplement is designed in accordance with ISO 11731 standards for the detection and enumeration of Legionella spp. from clinical and environmental samples, including hospital water systems, cooling towers, and other monitored environments.

Composition (per vial / per liter of medium)

• Glycine: 1.5 g / 3.0 g
• Vancomycin: 0.5 mg / 1.0 mg
• Polymyxin B: 40,000 IU / 80,000 IU
• Cycloheximide: 40.0 mg / 80.0 mg

Preparation Instructions

1. Reconstitute one vial with 5 mL sterile distilled water.
2. Mix until fully dissolved.
3. Add 1 mL of the reconstituted solution to 90 mL of autoclaved, cooled Legionella BCYE Agar Base.
4. Add 10 mL of reconstituted Legionella Growth Supplement (Ref. 80056) to complete the medium.
5. Mix thoroughly and dispense into Petri dishes.

Quality Control

• Legionella pneumophila WDCM 00107 → Good growth.
• Escherichia coli ATCC® 25922 → Partially to totally inhibited.
