Principle of the Test
Hippuric acid is hydrolyzed by the enzyme hippuricase into glycine and benzoic acid.
- Glycine reacts with ninhydrin, triggering a series of reactions involving hydrindantin, ammonia, and residual ninhydrin.
- This condensation reaction produces a purple color for a positive result, confirming hippurate hydrolysis.
- Negative reactions remain light gray.
