Liofilchem® Campylobacter Karmali Supplement is a selective lyophilized additive formulated to enrich Campylobacter Karmali Agar Base (Ref. 610200 / 620200) for the isolation of Campylobacter jejuni and Campylobacter coli from clinical and non-clinical specimens.
Developed according to Karmali et al. (1986), this supplement combines antimicrobial agents and growth factors to achieve optimal selectivity and aerotolerance for Campylobacter species. The resulting medium effectively suppresses competing flora, ensuring clear, reproducible isolation of target organisms under microaerophilic conditions at 42°C.
Key Features
- Highly selective supplement for Campylobacter jejuni and C. coli.
- Based on Karmali’s formulation, combining antibiotics and aerotolerance enhancers.
- Dual function: antimicrobial selectivity and improved Campylobacter growth.
- Each vial sufficient for 500 mL of medium.
- Freeze-dried and ready-to-reconstitute—ensures consistency and reliability.
- Validated under microaerobic incubation (5–10% O₂, 10% CO₂).
Composition
| Component | Per Vial | Per Litre of Medium | Function |
|---|---|---|---|
| Cefoperazone | 16.0 mg | 32.0 mg | Inhibits Gram-negative enteric bacilli |
| Vancomycin | 10.0 mg | 20.0 mg | Inhibits Gram-positive bacteria |
| Cycloheximide | 50.0 mg | 100.0 mg | Antifungal agent |
| Hemin | 16.0 mg | 32.0 mg | Enhances Campylobacter aerotolerance |
| Sodium Pyruvate | 50.0 mg | 100.0 mg | Detoxifies peroxides and supports growth |
Principle of the Method
The Campylobacter Karmali Supplement enhances selectivity and recovery of Campylobacter spp. through a combination of antimicrobials that inhibit competing organisms and growth factors that neutralize oxygen toxicity.
- Cefoperazone and Vancomycin: inhibit Gram-negative and Gram-positive contaminants.
- Cycloheximide: prevents fungal contamination.
- Hemin and Sodium Pyruvate: increase aerotolerance and support cell metabolism.
This combination creates a balanced environment that promotes rapid and efficient isolation of Campylobacter jejuni and C. coli from diverse sample types.
Preparation and Use
- Aseptically reconstitute the contents of one vial with 5 mL of sterile distilled water.
- Mix gently until fully dissolved.
- Add the entire reconstituted supplement to 500 mL of autoclaved Campylobacter Karmali Agar Base (45–50 °C).
- Mix carefully and pour into sterile Petri dishes.
- Incubate inoculated plates under microaerophilic atmosphere at 42°C for 18–48 hours.
Quality Control
| Organism | ATCC No. | Expected Result |
|---|---|---|
| Campylobacter jejuni | 33291 | Good growth |
| Escherichia coli | 25922 | Inhibited |
Appearance: whitish lyophilized plug, clear solution when reconstituted
Incubation conditions: 18–48 h at 42 °C, microaerophilic atmosphere
