Invitrogen™ D-Biotin is a high-purity, unconjugated form of biotin (also known as vitamin B7 or vitamin H) available in both liquid and powder formats. D-Biotin is widely used in molecular biology, immunoassays, affinity chromatography, and biochemical research due to its exceptionally high binding affinity to streptavidin, avidin, and NeutrAvidin™ proteins.

In its biologically active D-isomer form, biotin plays a key role in various enzyme cofactor functions and in labeling and detection workflows when paired with avidin-family binding proteins. It is a foundational reagent in biotin-streptavidin affinity systems commonly used in ELISA, blotting, and flow cytometry.

Key Features & Benefits

  • High-Affinity Binding
D-biotin forms one of the strongest known non-covalent bonds with streptavidin, avidin, and NeutrAvidin proteins—ideal for stable, high-sensitivity detection and capture assays.
  • Versatile Applications
Applicable in both chemical conjugation workflows and cellular labeling experiments, depending on format and protocol.
  • Two Convenient Formats
    • Liquid (50 mM) format for immediate use in assays without dissolution (Cat. No. B20656)
    • Powder format for custom formulation and long-term storage (Cat. No. B1595)
  • Unconjugated for Flexible Labeling
Enables researchers to modify or conjugate biotin to their molecule of interest using appropriate coupling chemistries (e.g., NHS ester, maleimide, click chemistry).
  • Compatible with Biotin-Binding Systems
Fully compatible with all biotin-binding proteins used in detection and purification systems (e.g., streptavidin magnetic beads, NeutrAvidin enzyme conjugates).

Applications

  • ELISA & Immunoassays
Use as a component in biotin-streptavidin signal amplification systems for target detection.
  • Protein & Nucleic Acid Labeling
Modify antibodies, enzymes, or oligonucleotides for immobilization or capture.
  • Affinity Purification
Create biotinylated ligands for capture on streptavidin-coated beads or plates.
  • Cell Surface & Intracellular Labeling
For selective detection or imaging of biomolecules in live or fixed cell systems.
  • Blotting Techniques
Biotinylated probes or antibodies for Western, Southern, and Northern blot detection protocols.
