Usage Guidelines
Preparation
- Suspend 13 g of powder in 1 liter of distilled or deionized water.
- Heat until completely dissolved.
- Dispense into sterile containers with or without Durham tubes.
- Autoclave at 121°C for 15 minutes.
Technique
- Pre-enrichment:
- Mix 25 g/mL of the sample with 225 mL of Lactose Broth and incubate at 35°C for 24 ± 2 hours.
- Enrichment:
- Transfer 1 mL of the pre-enrichment broth into a suitable enrichment broth and incubate.
- Selective Plating:
- Streak a loopful onto an appropriate agar plate.
Results Interpretation
- Gas production indicates lactose fermentation.
- Use selective plating and enrichment to isolate Salmonella and coliforms effectively.
