Medium for Cultivation of Yeasts and Moulds — USP/EP/JP Compliant
Liofilchem® Sabouraud Dextrose Broth (SDB) is a sterile, ready-to-use or dehydrated microbiological medium specifically designed for the cultivation and subculture of fungi, including yeasts and molds, from clinical and non-clinical materials. It is also widely used in the microbiological examination of non-sterile pharmaceutical, cosmetic, and personal care products, in accordance with harmonized pharmacopoeial standards (USP <62>, EP 2.6.13, JP 4.05).
This nutrient-rich medium provides the essential conditions for fungal recovery, including an acidic pH and high glucose concentration that support selective fungal growth while inhibiting most bacteria. It may also be supplemented with chloramphenicol for enhanced bacterial inhibition, especially in dermatophyte recovery.
Key Features and Benefits
- ✅ Pharmacopoeia-compliant (USP/EP/JP harmonized)
- ✅ Supports growth of dermatophytes, yeasts, and molds
- ✅ Liquid format ideal for direct inoculation or enrichment procedures
- ✅ Can be supplemented with antibiotics (e.g., chloramphenicol) for selective recovery
- ✅ Suitable for both clinical diagnostics and industrial microbiology applications
Typical Formula (per liter)
Component | Quantity |
---|---|
Pancreatic Digest of Casein | 5.0 g |
Peptic Digest of Animal Tissue | 5.0 g |
Dextrose | 20.0 g |
Final pH at 25 °C | 5.6 ± 0.2 |
Preparation Instructions (for dehydrated medium)
- Suspend 30 g of powder in 1 L of distilled or deionized water.
- Mix thoroughly and bring to boil with frequent agitation until fully dissolved.
- Dispense into appropriate containers.
- Sterilize by autoclaving at 121 °C for 15 minutes.
Test Procedure and Incubation
- Clinical use: Inoculate directly and incubate aerobically at 25 °C for 2–7 days.
-
Industrial use:
- For Candida albicans: Incubate at 30–35 °C for 3–5 days.
- For Aspergillus brasiliensis: Grow at 20–25 °C for 5–7 days.
- Subculture on Sabouraud Dextrose Agar (SDA) if needed.
Interpretation of Results
- Positive result: Turbidity indicates fungal growth.
- Negative result: No turbidity; no fungal growth observed.
Quality Control (QC)
