Liofilchem® Miconazole MCL Discs (10 µg) are high-quality antifungal susceptibility testing (AFST) discs intended for in vitro use with the Kirby-Bauer disk diffusion method. Each disc is impregnated with 10 micrograms of Miconazole, an imidazole-class antifungal agent with broad-spectrum activity against yeasts and dermatophytes, including Candida spp., Trichophyton spp., and Microsporum spp. These discs are manufactured in accordance with ISO 9001 and ISO 13485 standards, ensuring precision, sterility, and consistency for use in clinical diagnostics, dermatological microbiology, veterinary testing, and antifungal research.

    Key Features

    • Accurate dosage: Each disc contains 10 µg of Miconazole for standardized antifungal zone measurement.
    • Broad-spectrum activity: Effective against Candida albicans, Candida tropicalis, Trichophyton rubrum, Microsporum canis, and other dermatophytes.
    • Dermatology and mucosal application focus: Commonly used in testing fungi from skin, vaginal, and oral infections.
    • CLSI-validated method: Suitable for disk diffusion testing on specialized media for fungi.
    • Sterile and stable: Packaged in sealed cartridges or canisters with desiccant to maintain potency and prevent degradation.
    • Color-coded labeling: For easy disc identification in mycology labs.

    Applications and Use Cases

    1. Clinical and Dermatological Mycology

Used in routine diagnostics to assess susceptibility of:

    • Candida spp. in cases of oral, vaginal, or cutaneous candidiasis
    • Trichophyton, Epidermophyton, and Microsporum spp. from dermatophytosis (ringworm, athlete’s foot, jock itch)
    • Malassezia spp. in seborrheic dermatitis and tinea versicolor

    2. Veterinary Mycology

    • Test antifungal response in fungal skin and ear infections in domestic animals.

    3. Antifungal Stewardship and Resistance Monitoring

    • Track Miconazole resistance in dermatophytes and mucosal fungal isolates.
    • Support treatment optimization for topical antifungal formulations.

    4. Research and Pharmaceutical Development

    • Comparative testing with other imidazoles (e.g., Ketoconazole, Clotrimazole).
    • Evaluate new topical antifungal preparations or delivery vehicles.
    • Screen environmental or clinical fungal isolates for Miconazole efficacy.

    Use Instructions

    1. Prepare Sabouraud Dextrose Agar (SDA) or Mueller-Hinton Agar supplemented with 2% glucose and 0.5 µg/mL methylene blue (recommended for Candida spp.).
    2. Standardize the fungal inoculum to 0.5 McFarland turbidity using a spectrophotometer or visual comparator.
    3. Inoculate the agar surface evenly with a sterile swab.
    4. Apply one Miconazole disc (10 µg) using sterile forceps or a disc dispenser.
    5. Incubate:
      • For yeasts: 35 ± 2 °C for 24–48 hours
      • For dermatophytes: 28–30 °C for up to 5–7 days
    6. Measure the diameter of the inhibition zone in millimeters.
    7. Interpret using internal laboratory thresholds or published reference data, as CLSI and EUCAST breakpoints for Miconazole may be limited.

    Storage and Shelf Life

    • Storage Conditions: 2–8 °C (refrigerated), stored in sealed packaging with desiccant.
    • Shelf Life: 24–36 months from the date of manufacture under proper conditions.

Liofilchem® Miconazole MCL Discs (10 µg) offer a standardized, effective method for evaluating fungal susceptibility to a widely used topical antifungal. With broad activity against dermatophytes and yeasts, these discs are ideal for clinical, veterinary, and dermatological applications. Backed by ISO-certified quality and optimized for performance, they support effective antifungal therapy selection and resistance monitoring in routine and research settings.
