The Liofilchem™ Iodine Solution is a supplement used for the preparation of Tetrathionate Broth Base (Codes 610183, 620183). It facilitates the selective enrichment of Salmonella spp. from food and fecal specimens by inhibiting contaminating flora that lack the enzyme tetrathionate reductase.

Method Principle

• Tetrathionate Formation:
  • Upon addition of Iodine Solution, tetrathionate is formed in the medium, selectively inhibiting organisms that lack tetrathionate reductase and promoting the recovery of Salmonella spp.

Preparation Instructions

1. Prepare the Base Medium:
   • Follow instructions for preparing Tetrathionate Broth Base and cool to 45-50°C.
2. Add Iodine Solution:
   • Aseptically add 1 vial (10 mL) of Iodine Solution to 490 mL of medium.
   • Do not reheat after adding the supplement.
3. Mix and Dispense:
   • Mix thoroughly and pour into sterile containers.
4. Inoculate Quickly:
   • Use the complete medium within 2 hours of supplement addition.

Interpretation of Results

Quality Control

• Appearance:
  • Homogeneous dark brown liquid, clear.
• Microbiological Control:
  • Prepare the medium per label directions and inoculate with control strains.
  • Incubate at 35 ± 2°C for 18-24 hours.

Storage and Shelf Life

• Storage:
  • Store at 2–8°C, protected from light.
  • Can be stored or transported at 18–25°C for 4 days or 35–39°C for 48 hours without affecting performance.
• Shelf Life:
  • Valid until the expiry date indicated on the label.
