This low-concentration agar formulation enables visible detection of motile bacterial species via the diffusion of growth from the stab line in tube inoculations.

    Use Instructions

    Preparation:

    • Suspend 29.6 g of powder in 1 L of distilled or deionized water.
    • Heat to boiling with gentle shaking until completely dissolved.
    • Dispense into final containers.
    • Sterilize by autoclaving at 121°C for 15 minutes.

    Inoculation:

    • Inoculate by vertically stabbing a pure culture into the medium to more than half the depth of the tube.

    Incubation:

    • Primary: 36 ± 1°C for 24–48 hours in aerobic conditions.
    • Optional: Reincubate negative tubes at 25 ± 2°C for up to 5 days.

    Interpretation:

    • Motile organisms: Show diffuse or hazy growth radiating from the stab line.
    • Non-motile organisms: Growth confined to the stab line only.
